The enzyme-linked immunosorbent assay (ELISA) is currently used as an initial clinical screening of blood supplies. Students add simulated HIV antigen to wells in a microtiter plate. This is followed by washing and introduction of simulated antibodies of HIV positive or negative samples. A second antibody covalently linked to horseradish peroxidase enzyme is then added, preferentially binding to the first antibody of HIV positive samples. After washing, enzyme substrates are introduced, yielding a color reaction for HIV positive samples. You need: incubation oven, micropipets and tips, pipet bulbs for 1 mL pipet, glassware, gloves and goggles, and distilled or deionized water. Does not contain HIV or its components.
